Figure 1 Effect of cytarabine or melphalan±apraglutide on survival. Effect of cytarabine with or without apraglutide (3.3 mg/kg) on survival (group with daily dosing of apraglutide from day −4 not shown) (A); effect of cytarabine without apraglutide or with different doses of apraglutide on survival (B); and effect of melphalan with or without apraglutide 3.3 mg/kg on survival (high dose group 17.5 mg/kg apraglutide not shown) (C). (A) Kaplan-Meier curves. Animal survival was monitored daily. n=6–18 at time=0 for each treatment group. Apraglutide treatment initiated either before or concomitantly with the chemotherapy, showed significant difference relative to animals treated with cytarabine alone, as determined by log-rank (Mantel-Cox) test. When comparing to the vehicle/control group, survival of animals treated with cytarabine only was significantly different, as determined by log-rank (Mantel-Cox) test. No statistical difference was observed between the vehicle/control group and the groups that were treated with apraglutide. * P<0.0001. (B) Kaplan-Meier curves. GLP-2 receptor agonists (apraglutide, hGLP-2) were administered 4 days before and 5 days during cytarabine chemotherapy treatment. Animal survival was monitored daily. n=16 at time=0 for each treatment group; significant difference relative to vehicle/cytarabine-treated animals as determined by log-rank (Mantel-Cox) test. * P<0.05, # P<0.01. (C) Kaplan-Meier curves. Animal survival was monitored daily. n=10 at time=0 for each treatment group. When comparing to the vehicle/control group, survival of animals treated only with melphalan was significantly different as determined by log-rank (Mantel-Cox) test. No statistical difference was observed between the vehicle/control group or the melphalan-only group and the group treated with apraglutide 3.3 mg/kg. * P≤0.001. BID – twice daily; QD – once daily.
